Abstract
White matter, composed of axons ensheathed in myelin produced by mature oligodendrocytes, is essential for the rapid and efficient transmission of electrical impulses throughout the nervous system. The development of oligodendrocytes follows a tightly regulated lineage progression, beginning with neural progenitor cells (NPCs) that differentiate into oligodendrocyte progenitor cells (OPCs). These OPCs undergo proliferation, migration, and ultimately terminal differentiation into mature, myelinating oligodendrocytes. Disruptions at any stage of this developmental cascade—whether in lineage specification, proliferation, migration, or maturation—can impair myelination and contribute to the pathophysiology of neurodevelopmental disorders (NDDs). Therefore, identifying the precise stage at which oligodendrocyte lineage progression is altered is critical for elucidating the cellular and molecular mechanisms underlying NDDs.
