Abstract
Mycobacterium Tuberculosis (M.tb) remains one of the leading causes of death in the third world and current diagnostic methods take a long time to generate a result. A microwave mediated approach (using a TM 010 cylindrical cavity at 2.5GHz) to promote DNA release has been previously suggested as the first step (i.e. in the sample processing stage) of a rapid detection but was limited by the volume of sample it could process (up to 200µl). Here we propose a new cartridge geometry able to increase the sample volume up to 10ml, processed using the same cavity applicator. In a proof of principle study, we have used this cartridge to mediate significant DNA release in 10ml samples using Mycobacterium Smegmatis (M.smeg) and Mycobacterium Abscessus (M.Abs) cultures as a substitute for M.tb.
