Abstract

The inhibition of SPAK and OSR1 kinases binding to their upstream WNK kinases has been identified as a plausible strategy for inhibiting the WNK-activation of SPAK and OSR1 kinases. To facilitate the discovery of SPAK and OSR1 kinase inhibitors as potential therapeutics, we report the crystal structures of the highly conserved C-terminal domains of human SPAK and OSR1, which mediate the binding to, and activation, by their upstream WNK kinases.
