Abstract

The subject of this chapter is ADAMTS4. ADAMTS4 is a calcium-dependent metallo-endopeptidase homologous to the reprolysins. It is an ADAM protein that contains thrombospondin repeats. ADAMTS4 is an aggrecanase, cleaving the cartilage aggrecan core protein at Glu373. In addition, it cleaves other proteoglycans, including biglycan, brevican, versican, decorin, and fibromodulin, and glycoproteins such as fibronectin and transferrin. ADAMTS4 is inhibited by tissue inhibitor of metallopeptidase (TIMP)-3 (but not other TIMPs). ADAMTS4 is synthesized as a precursor and activated by a furin-like endopeptidase.
